Olaparib (LYNPARZA) is the ONLY PARP inhibitor recommended by NCCN Clinical Practice Guidelines in Oncology (NCCN Guidelines®) as both combination therapy and monotherapy for certain patients with mCRPC1-6

Indications NCCN Guidelines® for Prostate Cancer
Combination therapy in BRCAm mCRPC

Olaparib (LYNPARZA) is a poly (ADP-ribose) polymerase (PARP) inhibitor indicated in combination with abiraterone and prednisone or prednisolone (abi/pred) for the treatment of adult patients with deleterious or suspected deleterious BRCA-mutated (BRCAm) metastatic castration-resistant prostate cancer (mCRPC). Select patients for therapy based on an FDA-approved companion diagnostic for olaparib (LYNPARZA).

Pre-ARPI*

NCCN CATEGORY 1 Olaparib + abi/pred for BRCAm mCRPC

Monotherapy in HRRm mCRPC, including BRCAm

Olaparib (LYNPARZA) is a poly (ADP-ribose) polymerase (PARP) inhibitor indicated for the treatment of adult patients with deleterious or suspected deleterious germline or somatic homologous recombination repair (HRR) gene-mutated metastatic castration-resistant prostate cancer (mCRPC) who have progressed following prior treatment with enzalutamide or abiraterone. Select patients for therapy based on an FDA-approved companion diagnostic for olaparib (LYNPARZA).

Based upon high-level evidence (≥1 randomized phase 3 trials or high-quality, robust meta-analyses), there is uniform NCCN consensus (≥85% support of the Panel) that the intervention is appropriate (Category 1).

Based upon lower-level evidence, there is uniform NCCN consensus (≥85% support of the Panel) that the intervention is appropriate (Category 2A).
